Measurement of tissue impedance during electroporation

The Bachelor's thesis is devoted to the impedance of tissue in relation to the modern medical method electroporation. The Bachelor's thesis is divided into five chapters. The first three chapters describe the behaviour of the tissue and its impedance, whether during the passage of an electrical current or a phenomenon before and after the electroporation procedure. The fourth and fifth chapters examine those phenomena in the previous chapters on measurements made on animal and plant tissue.

This master´s thesis is dedicated to the ever-improving medical method - electroporation. This work is divided into six chapters. The first two chapters describe what electroporation entails and how it affects the tissue, how we can evaluate it and how much influence the change in tissue temperature has on measurements. The third chapter deals with the design of coaxial arrangement of electrodes - from the initial simulation to the physical product. In the fourth chapter, the measuring workplace is shown. The penultimate chapter is described and processed by individual experiments carried out on animal tissue. The last, sixth chapter summarizes the possible inaccuracies that arise in the measurement and therefore affect the results.

Regeneration abilities among vertebrates

Among vertebrates the ability to regenerate various body structure, from the cellular level to diverse internal organs or even the complex body structures (such as limb or tail), is unequally distributed across animal taxa. Regeneration is understood as the restoration of damaged or completely lost tissues, while the restored structure is more or less similar or even identical to the original structure. Regeneration is naturally often preceded by an encounter with a predator, when the animal is directly injured by the predator or an autotomy may occur as an anti-predator mechanism, which means that the animal voluntarily releases an appendage. Autotomy and following regeneration have many advantages but there can also appear some costs associated with these events which may have various consequences Masters in regeneration are considered to be mainly urodele amphibians who are able to perfectly regenerate various body structures (such as eye lens, limb or tail) and this ability persists throughout their lives, unlike anuran amphibians, in which the ability to regenerate after metamorphosis decreases significantly. During tissue processing, tissue machine occurs despite meticulous compliance with the procedures, exceptionally, damage to the tissues, which complicates their further processing into histological preparations. Determining where the error occurred is problematic, as damage is found at the very end of the processing cycle. This work deals with the clarification of possible causes, especially that in which step can the damage occur, what is caused, how they manifest and how to possibly prevent it.

Vliv triazinových pesticidů na ryby

Fish and crayfish are widely used as biological monitors of environmental levels of anthropogenic pollutants. The present thesis is a contribution to the assessment of the toxicity and effects of long-term effect of triazines on the different developmental stages of common carp (Cyprinus carpio L.) and adult red swamp crayfish (Procambarus clarkii). The carp was selected as a model fish due to its economic importance, e.g. carp farming contributes about 90% to total fish production in the Czech Republic. Crayfish are easily identified species representing given locality, they are widespread, and they provide a sufficient amount of tissue for individual biochemical and chemical analyses. The results of these studies provide further data on chronic exposure to triazines for consideration in risk assessment. We selected three active substances of triazine herbicides which are the most frequently detected in surface waters such as prometryne, simazine and terbutryn. The findings contribute to knowledge of the toxic potential of triazine herbicides to carp and crayfish at environmentally relevant concentrations in Czech rivers. There is a scarcity of information regarding the toxicity of triazines on freshwater organisms. During the tests we monitored several parameters: behaviour, mortality, biometric, haematological, biochemical blood, histopathology, oxidative stress and antioxidants. The data obtained from all tests performed during my thesis are very valuable for assessment and evaluation of long-term effects xenobiotics on aquatic organisms, especially fish and crayfish. In the future, I would extend the focus of my research for study of possible synergic or amplifying effect of mixtures triazines with other xenobiotics which are often found in the aquatic environment. This approach is recently accented as the aquatic environment is polluted by mixtures of different compounds. Therefore, more research is needed in order to clarify the more detailed effects of xenobiotics on non-target aquatic organisms.
